Role of HR-HPVs E6 and E7 Oncoproteins in Cervical Carcinogenesis

چکیده
The strategies that High Risk Human Papillomavirus (HR-HPVs) have developed in differentiating epithelial cells to reach a DNA-synthesis competent state are basically related to the overexpression of the E6 and E7 oncoproteins and the cellular regulatory pathways that are targeted by them. These are basically related to hallmarks of cancer, and include sustaining proliferative signals, the evasion of growth suppression and immune destruction, replicative immortality, inflammation, invasion, metastasis and angiogenesis, as well as genome instability, resisting cell death and deregulation of cellular energetics. In this minireview, we summarize some of the biological activities of HVP16 E6 and E7 oncoproteins in cervical carcinogenesis. From this, it is evident that cervical cancer may represent an important model to understand the different oncogenic mechanisms and processes involved not only in this neoplasia but also in the development of other human malignancies. Likewise, the miRNAs and cancer stem cellrelated markers expression could serve as novel molecular targets for the diagnosis and treatment of HPV positive

The Role of E6 Spliced Isoforms (E6*) in Human Papillomavirus-Induced Carcinogenesis
Persistent infections with High Risk Human Papillomaviruses (HR-HPVs) are the main cause of cervical cancer development. The E6 and E7 oncoproteins of HR-HPVs are derived from a polycistronic pre-mRNA transcribed from an HPV early promoter. Through alternative splicing, this pre-mRNA produces a variety of E6 spliced transcripts termed E6*. In pre-malignant lesions and HPV-related cancers, diffe...
متن کاملO-linked N-acetylglucosamine transferase promotes cervical cancer tumorigenesis through human papillomaviruses E6 and E7 oncogenes
O-linked N-acetylglucosamine (O-GlcNAc) transferase (OGT) increases O-GlcNAc modification (O-GlcNAcylation), and transcriptional co-regulator host cell factor 1 (HCF-1) is one of OGT targets. High-risk Human Papillomaviruses (HPVs) encode E6 and E7 oncoproteins, which promote cervical cancer. Here, we tested whether O-GlcNAc modification of HCF-1 affects HPV E6 and E7 expressions and tumorigene...
متن کاملE6^E7, a Novel Splice Isoform Protein of Human Papillomavirus 16, Stabilizes Viral E6 and E7 Oncoproteins via HSP90 and GRP78
UNLABELLED Transcripts of human papillomavirus 16 (HPV16) E6 and E7 oncogenes undergo alternative RNA splicing to produce multiple splice isoforms. However, the importance of these splice isoforms is poorly understood. Here we report a critical role of E6^E7, a novel isoform containing the 41 N-terminal amino acid (aa) residues of E6 and the 38 C-terminal aa residues of E7, in the regulation of...
متن کاملUpregulation of PIR gene expression induced by human papillomavirus E6 and E7 in epithelial oral and cervical cells
The hallmark of high-risk human papillomavirus (HR-HPV)-related carcinogenesis is E6 and E7 oncogene overexpression. The aim of this work was to characterize epithelial oral and cervical cancer cells that express HR-HPV E6 and E7 oncoproteins. Transcriptomic assay using DNA microarrays revealed that PIR gene expression was detected in oral cells in an HR-HPV E6/E7-dependent manner. In addition,...
متن کاملOverexpression of p16 Protein in Cervical Cancer in Thailand
The main cause of cervical cancer is a sexually transmitted infection by human papillomaviruses (HPVs) (Munoz and Bosch, 1992; Munoz et al., 1992; IARC, 1995; Walboomer et al., 1999). HPV infection has been detected in almost all preneoplastic and neoplastic lesions of the cervix (Zur Hausen, 1996). HPV contributes to neoplastic progression predominantly through the action of two viral oncoprot...
